A novel maternal mRNA storage compartment in mouse oocytes.
نویسنده
چکیده
Differentiation of a mouse oocyte into a two cell mouse embryo is controlled by the regulated translation of maternal mRNAs [1]. These mRNAs must be stored in the oocyte and then activated at the proper time in development. While advances in our understanding of mRNA storage and metabolism in somatic cells have been made, how mouse oocytes store and active maternal mRNAs is not well understood. In the current issue of Biology of Reproduction, Flemr and colleagues [2] address this issue and identify a new mRNA storage region in mouse oocytes called the subcortical RNP domain (SCRD).

I-52: Maternal mRNA Metabolism duringOocyte-to-Zygote Transition
Background: Maternal mRNA degradation is a selective process that occurs in waves corresponding to important developmental transitions such as resumption of meiosis, fertilization and zygotic genome activation. It has been demonstrated that the number, position, and combination of 3 UTR cis-acting elements interacting with trans-acting protein factors regulate translation and mRNA stability. Ou...
متن کاملTime Course of Degradation and Deadenylation of Maternal c-mos and Cyclin A2 mRNA during Early Development of One-Cell Embryo in Mouse
Early in the development of many animals, before transcription begins, any change in the pattern of protein synthesis is attributed to a change in the translational activity or stability of mRNA in the egg and early embryo. As a result, translational control is critical for a variety of developmental decisions, including oocyte maturation and initiation of preimplantation development. In this s...

Introduction & Objective: During oogenesis, mRNA is actively transcribed and accumulated in the growing oocytes, and then the transcription stops. Transcription silencing will continue during early embryonic stages at least up to the time when the embryonic genome is activated. Thus the earliest stages of embryogenesis in mammals and other animal species are depending on stored maternal RNAs an...
متن کاملA MAPK cascade couples maternal mRNA translation and degradation to meiotic cell cycle progression in mouse oocytes.
Mammalian oocyte maturation depends on the translational activation of stored maternal mRNAs upon meiotic resumption. Cytoplasmic polyadenylation element binding protein 1 (CPEB1) is a key oocyte factor that regulates maternal mRNA translation. However, the signal that triggers CPEB1 activation at the onset of mammalian oocyte maturation is not known. We provide evidence that a mitogen-activate...
متن کاملThe effect of ovary storage and in vitro maturation on mRNA levels in bovine oocytes; a possible impact of maternal ATP1A1 on blastocyst development in slaughterhouse-derived oocytes.
Since BSE testing of slaughtered cattle is obligatory in Japan, storage of ovaries at 15-20 C overnight in phosphate buffered saline has become a routine protocol in in vitro production (IVP) of cattle embryos. Ovary storage is known to reduce developmental competence of oocytes; however, its effects on oocyte gene expression have not been clarified yet. This study compared oocytes collected fr...
